Ammonium acetate is used for routine precipitation of nucleic acids, and is especially useful for reducing the coprecipitation of unwanted dNTPs and contaminating oligosaccharides. This solution is ready for use and requires no extra preparation. Note: Do not autoclave Ammonium Acetate Buffer. If precipitates form, warm solution to 37°C to resuspend. Applications: Routine Precipitation of DNA or RNA at Concentrations ≥20 ng/mL

This solution is ready for use and requires no extra preparation. Note: Do not autoclave Ammonium Acetate Buffer. If precipitates form, warm solution to 37°C to resuspend. Applications: Routine Precipitation of DNA or RNA at Concentrations ≥20 ng/mL
- 1. Use 5 M Ammonium Acetate to bring the final concentration of ammonium acetate in the solution to 2.5 M.
- 2. Add 2 volumes of 100% ethanol for DNA or 2.5 volumes of 100% ethanol for RNA.
- 3. Chill the solution at or below –20°C for ≥30 min.
- 4. Centrifuge at top speed in a microcentrifuge for 15 min.
- 5. Discard the supernatant and wash the pellet with ice cold 70–75% ethanol to remove residual salt.
- 6. Resuspend in nuclease-free water (Cat #AM9937) or 10 mM Tris, pH 7–8, 1 mM EDTA. Note: Ammonium acetate should not be used when the nucleic acid will be phosphorylated using T4 polynucleotide kinase, because this enzyme is inhibited by ammonium ions.
Description
Ambion® Molecular biology grade, 5 M Ammonium Acetate solution is supplied in one bottle containing 100 mL. The solution is certified RNase-free, economical, and ready-to-use. Due to the ubiquitous presence of RNases, manufacturing products for use with RNA is especially challenging. Ambion®'s nuclease-free reagents and buffers are manufactured in facilities specifically designed to prevent the introduction of nucleases. Highly sensitive RNase assays are performed at several different stages of the manufacturing process to ensure the highest quality. These reagents are rigorously tested for contaminating nonspecific endonuclease, exonuclease, and RNase activity.
