Target Cardiac Myosin Heavy Chain Target background Myosin is a highly conserved, ubiquitously expressed protein that hydrolyzes ATP, and this reaction provides the energy required for muscle contraction that interacts with actin to generate the force for cellular movements. It is a hexameric protein composed of four light chains and two heavy chains, each containing an actin-binding site and an ATP hydrolysis site. The heavy chains are encoded by the MYH gene family and were first isolated from a human fetal skeletal muscle and are the major determinant in the speed of contraction of skeletal muscle. Cardiac myosin exists as two isoforms in humans, Alpha-cardiac myosins and beta-cardiac myosins. These two isoforms are expressed in different amounts in the human heart. Beta-cardiac myosin s the predominant form during normal physiology while the alpha-isoform contributes for approximately 7% of the total myosin. Mutations of the MYH genes are associated with several different dilated and hypertrophic cardiomyopathies. Target alias CMHC Immunogen Human ventricular myosin Specificity This antibody recognizes alpha and beta type heavy chains of light meromyosin fragments (LMM) of cardiac myosin heavy chains. It also reacts with LMM fragments of cardiac myosin and beta heavy chain of slow human skeletal muscle. The antibody does not crossreact with human myosin light chains. Clone ID 3-48 Isotype IgG1 kappa Preservative None Format Purified with protein G, stored in PBS pH 7.4 and lyophilized. (Full IgG) Recommend starting dilution If reconstituted with deionized water with deionized in 100µL: WB: 1:500, IHC: 1:300. Optimal dilution has to be determined by the user. Limitations Research Use Only Storage Lyophilized antibodies can be kept at 4ºC for up to 3 months and should be kept at -20ºC for long-term storage (2 years). To avoid freeze-thaw cycles, reconstituted antibodies should be aliquoted before freezing for long-term (1 year) storage (-80ºC) or kept at 4ºC for short-term usage (2 months). For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made with the assay buffer. After the maximum long-term storage period (2 years lyophilized or 1 year reconstituted) antibodies should be tested in your assay with a standard sample to verify if you have noticed any decrease in their efficacy. To limit antibody loss or degradation, BSA (final concentration 1%) and sodium azide (final concentration 0.02%) can be added to the suggested first dilution. It is important to first verify if those preservatives are compatible with your assay.
