PSMA is a single-pass type II membrane protein that has both folate hydrolase and N-acetylated-alpha-linked-acidic dipeptidase activities. It exhibits a preference for tri-alpha-glutamate peptides. Its expression pattern is restricted mainly to the healthy prostate secretory-acinar epithelium and the plasma membrane of epithelial prostate carcinoma. In the brain, it modulates excitatory neurotransmission through the hydrolysis of the neuropeptide, N-aceylaspartylglutamate (NAAG) and release of glutamate. Clone 3F11 is shown to be highly specific for PSMA and does not exhibit any cross-reactivity with unrelated proteins. It binds to a linear epitope (aa 226-243) within the extracellular domain and is reported to be suitable for the detection of PSMA orthologs from some other species when denatured form of PSMA is used.
