Interleukin-17F (UniProt: Q7TNI7; also known as IL-17F) is encoded by the Il17f gene (Gene ID: 257630) in murine species. The IL-17 family of cytokines contains six members that share four conserved cysteine residues at their C-terminal region that participate in the formation of intermolecular disulfide linkages. Members of this family are associated with the pathogenesis of multiple autoimmune diseases including rheumatoid arthritis, multiple sclerosis, and inflammatory bowel diseases. IL-17F is an effector cytokine of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity. It is a disulfide-linked homodimeric protein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to produce mature secreted protein (aa 29-161). It can also form a heterodimer with IL-17A. Its actions are mediated via interaction with IL-17RA and IL-17RC receptors with 2:1 binding stoichiometry. It is primarily involved in host defense against extracellular bacteria and fungi by inducing neutrophilic inflammation. As signature effector cytokine of T-helper 17 cells (Th17), it induces neutrophil activation and recruitment at infection and inflammatory sites. It is also known to stimulate the production of antimicrobial β-defensins by mucosal epithelial cells thereby limiting the entry of microbes through the epithelial barriers.
