Glycerol-3-phosphate acyltransferase 4 (UniProt: Q86UL3; also known as EC:2.3.1.15, 1-acylglycerol-3-phosphate O-acyltransferase 6, 1-AGP acyltransferase 6; 1-AGPAT 6, Acyl-CoA:glycerol-3-phosphate acyltransferase 4, Lysophosphatidic acid acyltransferase zeta, LPAAT-zeta, Testis spermatogenesis apoptosis-related protein 7, TSARG7) is encoded by the GPAT4 (also known as AGPAT6, TSARG7, UNQ551/PRO1108) gene (Gene ID: 137954) in human. GPAT4 is a member of the 1-acylglycerol-3-phosphate O-acyltransferase family that plays a role in triglyceride biosynthesis in several tissues. It is a rate-limiting enzyme that converts glycerol-3-phosphate (G3P) to 1-acyl-sn-glycerol-3-phosphate by incorporating an acyl moiety at the sn-1 position of the glycerol backbone. It is shown to be active against both saturated and unsaturated long-chain fatty acyl-CoAs. Its HXXXXD motif (aa 248-253) is reported to be essential for its acyltransferase activity and may constitute the binding site for the phosphate moiety of the glycerol-3-phosphate. By increasing triglyceride synthesis, it protects cells against lipotoxicity induced by high levels of saturated fatty acids. (Ref.: Piccolis, M., et al. (2019). Mol.
